چکیده

The Present paper deals with the herbicide Butachlor (5,10 ,20,40 and 80ppm)-induced changes in physiological and biochemical parameters related to photosynthesis and defense systems in paddy field cyanobacterium Plectonema boryanum grown under laboratory conditions. Growth and photosynthetic pigments, i.e., chlorophyll a and carotenoids were adversely affected by Butachlor treatment and the inhibition was found to be dose dependent. The toxic effect of Butachlor was more pronounced protein; however, a considerable reduction in chlorophyll a, and carotenoids was also noticed. Furthermore, Butachlor with increasing doses accelerated the formation of active oxygen species, i.e., O and HO, in cells progressively. As a consequence of active oxygen species (AOS) generation in Butachlor -treated cells, the activity of superoxide dismutase (SOD) and peroxidase (POD) was enhanced considerably. Besides the accelerated action of enzymatic defense systems, Protein damage also showed an increasing trend with the rising concentration of Butachlor (5, 10, 20, 40 and 80 ppm).

A mobilizable shuttle vector for the cyanobacterium Plectonema boryanum

Plasmid pSUP5011 contains a pMBl origin of replication, an origin of transfer (oriT), and genes encoding resistance to the antibiotics ampicillin, chloramphenicol and kanamycin. pSUP5011 was conjugally mobilized from Escherichia coli into the non-heterocystous, filamentous, nitrogen-fixing cyanobacterium Plectonema boryanum UTEX 594 in the presence of a helper plasmid, RP4. Transconjugant cyano...

Effects of pesticides on cyanobacterium Plectonema boryanum and cyanophage LPP-1.

Cyanobacterium Plectonema boryanum IU 594 and cyanophage LPP-1 were used as indicator organisms in a bioassay of 16 pesticides. Experiments such as spot tests, disk assays, growth curves, and one-step growth experiments were used to examine the effects of pesticides on the host and virus. Also, experiments were done in which host or virus was incubated in pesticide solutions and then assayed fo...

Differential expression of photosynthesis and nitrogen fixation genes in the cyanobacterium Plectonema boryanum.

The filamentous non-heterocystous cyanobacterium Plectonema boryanum fixes dinitrogen at a high rate during microaerobic growth in continuous illumination by temporal separation of oxygen-evolving photosynthesis and oxygen-sensitive dinitrogen fixation. The onset of nitrogen fixation is preceded by a depression in photosynthesis that establishes a sufficiently low level of dissolved oxygen in t...

A novel nitrite reductase gene from the cyanobacterium Plectonema boryanum.

The gene (nirA) for nitrite reductase was cloned from the nonheterocystous, filamentous cyanobacterium Plectonema boryanum. The predicted protein consists of 654 amino acids and has a calculated molecular weight of 72,135. The deduced amino acid sequence from positions 1 to 511 is strongly similar to the entire sequence of the ferredoxin-dependent nitrite reductases from other phototrophs, whil...

Differential operation of dual protochlorophyllide reductases for chlorophyll biosynthesis in response to environmental oxygen levels in the cyanobacterium Leptolyngbya boryana.

Most oxygenic phototrophs, including cyanobacteria, have two structurally unrelated protochlorophyllide (Pchlide) reductases in the penultimate step of chlorophyll biosynthesis. One is light-dependent Pchlide reductase (LPOR) and the other is dark-operative Pchlide reductase (DPOR), a nitrogenase-like enzyme assumed to be sensitive to oxygen. Very few studies have been conducted on how oxygen-s...
